Having created a diagram showing all the people that I want for a particular purpose is it possible to create a list of all the people that appear in the diagram?

You can select all the boxes shown in the diagram and then use the option on the Edit menu to Add to Named List.... You can then report the list or use the list as a filter in a query.

Thanks Peter. Is there a way to select all 100 or so boxes without doing them one by one?

You can click the View Whole Diagram button (looks like  a picture of the world - also via the Zoom option of the Diagram menu) and then use the mouse to drag a rectangular box around the outside of the whole tree.  You need to ensure all required boxes are completely inside the dragged rectangle.
